Comparison of LIVE/DEAD¢ç BacLight¢â Bacterial Viability Test and alamarBlue¢ç Method for Enumeration of Live and Dead Bacteria for Oral Bacterial Species

Abstract
LIVE/DEAD¢ç BacLight¢â and alamarBlue¢ç are fluorescentmaterials used for the enumeration of live and deadbacteria. LIVE/DEAD¢ç BacLight¢â is generally used forconfocal microscopy applications to differentiate live fromdead bacteria in a biofilm or planktonic state. AlamarBlue¢çhas also been used widely to assay live and dead bacteria in aplanktonic state. Whilst these materials are successfully utilizedin experiments to discriminate live from dead bacteriafor several species of bacteria, the application of these techniquesto oral bacteria is limited to the use of LIVE/DEAD¢ç BacLight¢â in biofilm studies. In our present study,we assessed whether these two methods could enumerate liveand dead oral bacterial species in a planktonic state. Wetested the reagents on Streptococcus mutans, Streptococcussobrinus, Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitansand Enterococcus faecalis and found thatonly LIVE/DEAD¢ç BacLight¢â could differentiate live fromdead cells for all five of these oral strains. AlamarBlue¢ç wasnot effective in this regard for P. gingivalis or A. actinomycetemcomitans.In addition, the differentiation of liveand dead bacterial cells by alamarBlue¢ç could not be performedfor concentrations lower than 2 ¡¿ 106 cells/ml. Ourdata thus indicate that LIVE/DEAD¢ç BacLight¢â is a moreeffective reagent for this analysis.
